RESUMO

OBJECTIVES: The splicing factor transformer-2 homolog beta (Tra2ß) plays a pivotal role in various cancers. Nonetheless, its role in oral squamous cell carcinoma (OSCC) has not been comprehensively explored. This study sought to discern the influence of Tra2ß on OSCC and its underlying mechanisms. MATERIALS AND METHODS: We assessed Tra2ß expression in OSCC utilizing immunohistochemistry, qRT-PCR, and western blotting techniques. siRNA transfection was used to silence Tra2ß. Whole transcriptome RNA sequencing (RNA-seq) analysis was carried out to reveal the alternative splicing (AS) events. KEGG pathway analysis enriched the related pathways. Colony formation, transwell, wound healing, and Annexin V-FITC/PI were employed to appraise the consequences of Tra2ß silencing on OSCC. RESULTS: Tra2ß was highly expressed in both OSCC tissues and cell lines. Knockdown of Tra2ß-regulated AS events with skipped exon (SE) accounts for the highest proportion. Meanwhile, downregulation of Tra2ß reduced cell proliferation, migration, and invasion, however increasing cell apoptosis. Moreover, Wnt signaling pathway involved in the function of Tra2ß knockdown which was demonstrated directly by a discernible reduction in the expression of GSK3/ß-catenin signaling axis. CONCLUSIONS: These findings suggest that knockdown of Tra2ß may exert anti-tumor effects through the GSK3/ß-catenin signaling pathway in OSCC.

2D van der Waals (vdW) magnets open landmark horizons in the development of innovative spintronic device architectures. However, their fabrication with large scale poses challenges due to high synthesis temperatures (>500 °C) and difficulties in integrating them with standard complementary metal-oxide semiconductor (CMOS) technology on amorphous substrates such as silicon oxide (SiO2 ) and silicon nitride (SiNx ). Here, a seeded growth technique for crystallizing CrTe2 films on amorphous SiNx /Si and SiO2 /Si substrates with a low thermal budget is presented. This fabrication process optimizes large-scale, granular atomic layers on amorphous substrates, yielding a substantial coercivity of 11.5 kilo-oersted, attributed to weak intergranular exchange coupling. Field-driven Néel-type stripe domain dynamics explain the amplified coercivity. Moreover, the granular CrTe2 devices on Si wafers display significantly enhanced magnetoresistance, more than doubling that of single-crystalline counterparts. Current-assisted magnetization switching, enabled by a substantial spin-orbit torque with a large spin Hall angle (85) and spin Hall conductivity (1.02 × 107 ℏ/2e Ω⁻¹ m⁻¹), is also demonstrated. These observations underscore the proficiency in manipulating crystallinity within integrated 2D magnetic films on Si wafers, paving the way for large-scale batch manufacturing of practical magnetoelectronic and spintronic devices, heralding a new era of technological innovation.

The mechanism design theory can be applied not only in the economy but also in many fields, such as politics and military affairs, which has important practical and strategic significance for countries in the period of system innovation and transformation. As Nobel Laureate Paul said, the complexity of the real economy makes it difficult for "Unorganized Markets" to ensure supply-demand balance and the efficient allocation of resources. When traditional economic theory cannot explain and calculate the complex scenes of reality, we require a high-performance computing solution based on traditional theory to evaluate the mechanisms, meanwhile, get better social welfare. The mechanism design theory is undoubtedly the best option. Different from other existing works, which are based on the theoretical exploration of optimal solutions or single perspective analysis of scenarios, this paper focuses on the more real and complex markets. It explores to discover the common difficulties and feasible solutions for the applications. Firstly, we review the history of traditional mechanism design and algorithm mechanism design. Subsequently, we present the main challenges in designing the actual data-driven market mechanisms, including the inherent challenges in the mechanism design theory, the challenges brought by new markets and the common challenges faced by both. In addition, we also comb and discuss theoretical support and computer-aided methods in detail. This paper guides cross-disciplinary researchers who wish to explore the resource allocation problem in real markets for the first time and offers a different perspective for researchers struggling to solve complex social problems. Finally, we discuss and propose new ideas and look to the future.

Anthocyanins have great health benefits, especially malvidin. Vitis amurensis Rupr are rich in malvidin, and malvidin-3-O-glucoside (Mv3G) monomer is the most abundant. However, natural anthocyanins are unstable, which limits their wide application in the food field. Soybean insoluble dietary fiber (SIDF) has high stability, and it can be used as an inert substrate to construct a stable system, which may improve the stability of anthocyanins. The optimal condition to construct a stable system of SIDF and Mv3G at pH 3.0 was determined by an orthogonal experiment. The results indicated that SIDF effectively improved the stability of Mv3G under different pH values (1.0~7.0), high temperature (100 °C for 100 min), and sunlight (20 ± 2 °C for 30 d) conditions. The absorption peak intensity of the UV-VIS spectrum of SIDF-Mv3G was enhanced, which indicated that there was interaction between SIDF and Mv3G. Fourier transform infrared spectroscopy analyses revealed that the -OH stretching vibration peak of SIDF-Mv3G was changed, which indicated that the interaction between SIDF and Mv3G was due to hydrogen bonding. X-ray diffraction analysis showed that the crystalline morphology of SIDF was opened, which was combined with Mv3G, and SIDF made Mv3G change to a more stable state. Scanning electron microscope analysis showed that SIDF and Mv3G were closely combined to form an inclusion complex. Overall, this study provides valuable information for enhancing the color stability of anthocyanins, which will further expand the application of anthocyanins in the food field.

Anthocyanins are abundant in purple corn and beneficial to human health. Soybean protein isolate-7s (SPI-7s) could enhance the stability of anthocyanins. The stable system of soybean protein isolate-7s and delphinidin-3-O-glucoside complex (SPI-7s-D3G) was optimized using the Box-Behnken design at pH 2.8 and pH 6.8. Under the condition of pH 2.8, SPI-7s effectively improved the sunlight-thermal stabilities of delphinidin-3-O-glucoside (D3G). The thermal degradation of D3G conformed to the first order kinetics within 100 min, the negative enthalpy value and positive entropy value indicated that interaction was caused by electrostatic interaction, and the negative Gibbs free energy value reflected a spontaneous interaction between SPI-7s and D3G. The interaction of SPI-7s-D3G was evaluated by ultraviolet visible spectroscopy, circular dichroism spectroscopy and fluorescence spectroscopy. The results showed that the maximum absorption peak was redshifted with increasing the α-helix content and decreasing the ß-sheet contents, and D3G quenched the intrinsic fluorescence of SPI-7s by static quenching. There was one binding site in the SPI-7s and D3G stable system. The secondary structure of SPI-7s had changed and the complex was more stable. The stabilized SPI-7s-D3G will have broad application prospects in functional foods.

Preeclampsia affects 2-8% of all pregnancies, and it is associated with significant perinatal mortality and morbidities including preterm birth and small for gestational age. We examined whether plasma fatty acid binding protein 4 (FABP4) concentrations are associated with risk of later preeclampsia development. From March 2015 to May 2016, serum FABP4 was measured in 1486 women in early pregnancy. The relationship between the levels of FABP4 and preeclampsia were evaluated using univariate and multivariate regression analysis. The median plasma concentration of FABP4 at the first prenatal visit was significantly higher in women in whom preeclampsia developed later compared with those in whom it did not (P < 0.001). For each 1 unit increase in FABP4 plasma concentration, the unadjusted and adjusted risk of preeclampsia increased by 8% (odds ratio (OR): 1.08; 95% confidence interval [CI]: 1.05-1.12) and 4% (1.04; 95% CI: 1.02-1.07), respectively. The addition of FABP4 to established risk factors significantly improved net reclassification improvement. Increased FABP4 at the first prenatal visit of gestation independently predicted preeclampsia and significantly improved reclassification and discrimination. This information is important to guide public health efforts in preeclampsia prevention.

Purpose: To evaluate the safety and efficacy of Enterprise stent in the treatment of symptomatic complex intracranial atherosclerotic stenosis. Methods and materials: 20 complex cases among 53 patients with symptomatic ischemic stroke who were treated with balloon angioplasty and enterprise stents in Department of Neuro-interventional Radiology, The First Affiliated Hospital of Zhengzhou University from Jan. 2014 to Dec. 2017 were retrospectively enrolled in this study. Diagnostic criteria for complex intracranial atherosclerotic stenosis were as follows: 1) intracranial atherosclerotic stenosis was ≥70% confirmed by digital subtraction angiography (DSA) using the formulas described by the Warfarin Aspirin Symptomatic Intracranial Disease (WASID) method; 2) length of lesion was >10 mm. Basic characteristics of target lesions, technical success rate, perioperative safety, follow-up outcomes were investigated. Results: 20 patients were enrolled in this study, including 15 males and 5 females from 44 to 70 years old with an average age of 57.20 ± 9.25. 20 lesions were treated with 20 enterprise stents. The average preoperative and postoperative residual stenosis was reduced from (77.45 ± 8.44)% to (24.89 ± 16.61)%. The successful rate of operation was 100%. Among the perioperative complications, only 1 case (5%) experienced perforating branch event. The average clinical follow-up period was 13.15 ± 11.33 months (time range: 5-38). There were no ischemic events, no bleeding events and no various causes of death during the follow-up period. 8 lesions (40.0%) underwent DSA follow-up examinations and 12 lesions (60.0%) were checked by CT angiography during the follow-up period. 3 lesions (15.0%) developed ISR without any cerebral ischemia symptoms. Conclusion: This retrospective, single-center study suggests that enterprise stent is effective in the treatment of symptomatic complex intracranial atherosclerotic stenosis with less perioperative complications. Prospective, multicenter, randomized controlled trials are expected.
